Title: A Real Time PCR quantitative assay for Hepatitis B DNA virus in the blood sera of cornea donors with positive serology for Hepatitis B core antibody

Purpose:

The first line of blood screening is for hepatitis virus B (HBV). Tissue transplantation of all cornea donors with positive serology Hb core Ab weren’t performed and they was returned. With increasing suspected cases of HBV infection, testing with high accuracy in donors sera was necessary. We used Real Time PCR technique for detection of HBV in hepatitis B core antibody positive samples (HB core Ab+), since it is a method with the advantages of high sensitivity, broader linearity and less carryover contamination risks.

Methods:

50 serum positive Hb core Ab samples (with Hbs Ag negative) provided by eye bank of Iran. DNA was extracted from samples and analyzed by Real Time PCR technique.

Results:

50 samples with positive serology for Hb core Ab were analyzed and only 5 samples were positive for HBV.

Conclusion:

Our results showed that 90% of Hb core Ab positive corneal samples (for HBV) were completely safe and the results of this method, economically and ethically is important for rejection or transplantation of corneas.
